In order to find an 'A' holder, do I have to look at every player card?

If you go to your Advanced Depth Chart and select Holder, you'll be able to see every option on your team for that position and rank them.

if you're looking on the wire or in a draft though, you do need to open the individual cards. look for guys with good hands and exe ratings.

Holder used to be listed along with other Best Roles but a lot of people didn't like seeing their QB's showing a Best Role of Holder so it was removed. dirtdevil is right, Hands and Execution make for good holders. Most of your best holders are going to be QB's, Punters, and the occasional WR.
